Genetic Causes of Schizophrenia Increase Risks for Rare Developmental Disorders

Researchers from the Wellcome Trust Sanger Institute found that common genetic factors for common disorders can also be responsible for rare neurodevelopmental disorders. The study finds that an individual’s entire genetic make-up can alter the effects that rare damaging variants have for causing neurodevelopmental problems. This means simple genetic variants that cause common conditions such as schizophrenia can increase the risks of developing rare, damaging disorders.

The study was titled Common Genetic Variants Contribute to Risk of Rare Severe Neurodevelopmental Disorders and published in the journal Nature.

Damaged Genetic Variants Responsible For Autism and Other Neurodevelopmental Disorders

The main takeaway from the Deciphering Developmental Disorders (DDD) study is that rare genetic disorders are more complex than earlier believed.

According to scientists, thousands of newborn babies do not develop normally due to genetic factors. When children with problematic genetic variants grow up, they develop neurodevelopmental disorders that could result in learning disabilities or language development. Some could develop autism and other related neurodisorders.

To this extent, the DDD study set out to properly find diagnoses for children with yet-to-be known developmental conditions; and to fully understand the causes of the disorders.

Before this time, medical scientists used to think that common genetic variants determined commonly seen disorders such as diabetes and heart diseases. And that chronic mutations in single genes caused rare disorders. But this new study indicated otherwise. The study further confirms to the authors that different patients with damaging mutations in the same gene do not present the same symptoms. But the DDD researchers tried to know why this is so.

Common Genetic Variants Could Influence Changes in Rare Genetic Variants

For the study, the genomes of about 7,000 children with various neurodevelopmental disorders were analyzed and compared with those of healthy kids. The research team applied the Genome Wide Association Studies (GWAS) to compare up to four million common genetic variants linked to uncommon developmental problems. Then they found same common variants influenced the risks for uncommon neuro-disorders.

According to Mari Niemi of the Wellcome Sanger Institute, the common genetic variants can influence unusual changes during brain development, causing undesired developmental problems. Dr. Hilary Martin said other researchers and clinicians will now understand the interconnectedness between common and rare genetic diseases. He said this is the first ever large-scale study of its kind.

“As we learn more about the genetic causes of both rare and common diseases, it is increasingly clear that many different types of genetic variation play important roles in all cases,” said Dr. Jeffrey Barrett, senior author of the study.
